Enzymes are critical reagents for cell isolation or recovery of cells after in vitro culture. The limited choice of GMP-Grade commercial enzymes for these applications presents a hurdle to developers who need an alternative source of a critical enzyme. VitaCyte purifies and characterizes Clostridium histolyticum collagenases and other neutral proteases suitable for various applications. The case study below highlights how VitaCyte applied this expertise to purify and characterize a neutral protease from Aspergillus melleus used for isolating human amniotic epithelial cells (AEC). Company X licensed a university patent that described human AECs' application for cell therapy or their culture supernatants as biotherapeutic products. After licensing, the Company replaced trypsin with Sigma Protease XXIII from A. melleus, improving the attachment of AECs to culture vessels and increasing the secretion of bioactive factors into the culture supernatant. VitaCyte purified the predominant neutral protease from the crude Protease XXIII product, enabling it to be used as an ancillary material to isolate the AECs from amniotic membranes. Within six months, VitaCyte provided a sufficient amount of purified enzyme to show that productivity of the cell isolation process improved > 3x when the purified protease was used in place of Protease XXIII.
