Bio-Rad Presentation: Maximizing droplet digital PCR's Potential for Gene Therapy: Genome Integrity & Capsid Ratio Analysis
Thursday, September 5, 2024 10:15 AM - 10:45 AM
Global Showcase Presentation
Presentor:
  • Caroline Weydert, Bio-Rad Laboratories NV, Belgium
Digital PCR has become the standard for quantifying viral genomic titers and (vector) copy numbers for cell and gene therapeutic products. Also, residual host cell DNA, residual plasmid, and other biosafety parameters can be assessed using this technology. Beyond these common applications, genome integrity and AAV empty/full capsid ratios can be measured. Several parameters and integrity calculations are key for the accuracy of these applications.

Session Objectives:
  • ddPCR can generate genome titer data and estimate genome integrity estimates
  • ddPCR can determine empty/full capsid ratios without the need for ELISA
  • Linkage is used for these methods
  • Linkage is calculated
  • Multiple parameters, including sample prep, impact linkage data
  • ddPCR can be used for additional analytical applications
